Glutathione&©|160;(GSH) is an important&©|160;antioxidant&©|160;in plants, animals, fungi, and some bacteria and archaea, preventing damage to importantcellular&©|160;components caused by&©|160;reactive oxygen species&©|160;such as&©|160;free radicals,&©|160;peroxides,&©|160;lipid peroxides&©|160;and&©|160;heavy metals. It is atripeptide&©|160;with a&©|160;gamma peptide linkage&©|160;between the&©|160;carboxyl&©|160;group of the&©|160;glutamate&©|160;side-chain&©|160;and the&©|160;amine group&©|160;of&©|160;cysteine&©|160;(which is attached by normal peptide linkage to a&©|160;glycine).
Thiol&©|160;groups are&©|160;reducing agents, existing at a concentration around 5&©|160;mM&©|160;in&©|160;animal&©|160;cells. Glutathione reduces&©|160;disulfide bonds&©|160;formed within&©|160;cytoplasmic&©|160;proteins&©|160;to&©|160;cysteines&©|160;by serving as an&©|160;electron&©|160;donor. In the process, glutathione is converted to its oxidized form,glutathione disulfide&©|160;(GSSG), also called&©|160;L-(¨C)-glutathione.
Once oxidized, glutathione can be reduced back by glutathione reductase, using&©|160;NADPH&©|160;as an electron donor. The ratio of reduced glutathione to oxidized glutathione within cells is often used as a measure of cellular toxicity. |
